  • What is Sermorelin?

    Sermorelin is a peptide, meaning it is made of amino acids bonded together via peptide bonds. More specifically, it is made of 29 amino acids, which are identical to the shortest functional sequence of GHRH that is sufficient to stimulate GH synthesis.

    This is why sermorelin is also commonly referred to as GRF 1-29 (growth hormone-releasing factor 1-29).

    Sermorelin mimics the naturally occurring GHRH and stimulates the release of GH in a similar manner. When released, GH plays a major role in the growth of children as well as regulating the metabolism and body composition of adults.

    The peptide was developed by the German pharmaceutical company Merck Serono (EMD Serono in the United States and Canada.

    It was first approved in 1990 by the US Food and Drug Administration (FDA) as an intravenous formulation for the diagnosis of growth hormone deficiency (GHD) under the brand name Geref.

    In 1997, Geref (sermorelin) was also approved for subcutaneous injection therapy in children with growth failure due to GHD and other causes.

    The peptide has also shown favorable effects in adults with and without GHD when applied as either once-daily or twice-daily injections.

  • How Does Sermorelin Work?

    As an analog to GHRH, sermorelin can induce a significant increase in serum GH levels. This, in turn, upregulates the production of GH’s main anabolic mediator – insulin-like growth factor-1 (IGF-1).

    More specifically, sermorelin appears to stimulate an increase in GH levels within several minutes of administration. Due to the 10-minute half-life of sermorelin, the increase in GH is relatively short-lived, lasting for about 2 hours after an injection.

    Nevertheless, trials lasting up to 6 weeks report that even once-daily administration is sufficient to increase 12-hour mean GH levels (as measured by the area under the curve) by 82% from baseline.

    Twice-daily administration (a total of 2mg/daily) has also been shown to boost IGF-1 levels by over 21% in 6 out of the 10 elderly study participants within just 2 weeks.

    Longer trials, lasting up to 16 weeks, report that sermorelin at a dose of 10 mcg/kg of body weight can lead to a 107% increase in mean GH and a 28% increase in mean IGF-1 levels, on average, amongst elderly men.

  • How to use Sermorelin

    Sermorelin is usually administered once or twice daily in a total daily dose of 10-25 mcg/kg of body weight. Most trials in adults also apply anywhere between 1-2 mg of the peptide per day.

    Due to the short half-life of the peptide, it can be taken as either a single or twice daily injection. Studies involving twice-daily injections report that the evening ones are more effective than the injections in the morning.

    Thus, once-daily injections should be administered only in the evening. Twice-daily injections can be administered in the evening before bed and early in the morning, a couple of hours before breakfast.

    The peptide is usually sold as a lyophilized powder that requires reconstitution before use. Prior to reconstitution, the peptide is stable and can be stored in a dark, cool, dry place for extended periods of time.

    Once reconstituted, the shelf-life of the peptide will depend on the solvent used for the process. Bacteriostatic water, which contains a 0.9% benzyl alcohol as a preservative, is the recommended choice as it extends the shelf-life of sermorelin and other peptides by 28 days when refrigerated at 36 to 46 degrees Fahrenheit (2 to 8 degrees Celsius).

    Once reconstituted, sermorelin must be injected only subcutaneously. This can happen at specific injection sites, which are considered suitable for subcutaneous injections, such as:

    • The abdominal area, about 2 inches away from the navel
    • Outer upper arms
    • Front outer thighs

    Before administering the reconstituted peptide, make sure that you also have sterile needles, syringes, alcohol pads, and a sharps container available.

    Then you can follow the following step-by-step guide for sermorelin injections:

    • Perform hand hygiene by washing with soap and water, then disinfect your hands with a hand sanitizer. Then, put on a pair of sterile gloves.
    • Disinfect the top of the vial using an alcohol swab by wiping it in one direction.
      With a sterile syringe, extract the required amount of peptide solution from the vial. Avoid shaking or tapping the syringe to prevent peptide damage.
    • Prepare the injection site by cleaning the area with an alcohol swab. Let the alcohol evaporate.
    • Pinch the skin at the site with your non-dominant hand to form a fold.
    • With your dominant hand, hold the syringe with the needle bevel up and insert it fully into the skinfold at a 45-degree angle.
    • Gently press the plunger of the syringe to inject the solution into the skin.
    • Wait for a few seconds after the injection, then smoothly withdraw the needle at the same angle it was inserted.
    • Dispose of the needle and syringe immediately in a designated sharps container.
    • Apply a sterile alcohol swab to the injection site, applying gentle pressure to stop any bleeding.

FAQs

How Long Does Sermorelin Stay In Your System?

Sermorelin has a half-life of about 10 minutes. Considering that it takes 4-5 half-lives on average for a medication to completely leave the plasma, the peptide should not be detectable in serum after an hour post-administration. Yet, sermorelin may be detected for up to 2 hours after administration in a blood test. There is no data on how long it is detectable in urine, but considering the aforementioned study, it may be detectable for a few hours.

What Happens When You Stop Taking Sermorelin?

Sermorelin does not pose a risk of psychological or physical addiction, and stopping it cold turkey does not cause withdrawal. Further, it does not suppress the function of the pituitary gland, so GH and IGF-1 return to pretreatment levels after stopping sermorelin.

Does Sermorelin Affect Other Hormones?

As a GHRH-analog, sermorelin is highly selective and does not affect the synthesis of other hormones. Furthermore, sermorelin does not have a negative effect on insulin sensitivity or other aspects of endocrine health. The majority of studies do not report any effect on testosterone levels.
